GPCRs represent the largest family of signaling proteins targeted by many clinically used drugs. G-protein-coupled receptors (GPCRs) constitute a large and diverse family of proteins whose primary function is to transduce extracellular stimuli into intracellular signals. They are among the largest and most diverse protein families in mammalian genomes. The binding of a signal molecule by the extracellular part of the G-protein linked receptor causes the cytosolic tail of the receptor to interact with, and alter the conformation of, a G-protein. This has two consequences: First, the alpha subunit of the G- protein loses its GDP and binds a GTP instead. G-protein-coupled receptors (GPCR) are the most diverse group of membrane receptors in eukaryotes.
